TNF-a-induced activation of CAM expression argues for the major target of action of isoliquiritigenin possibly mediated via the transcriptional mechanisms. Licorice root extract contains many different subclasses of flavonoids, for instance isoflavans, isoflavones, and chalcones 1214 ; . Isoliquiritigenin, a flavonoid with a simple chalcone structure 4, 29, 49-trihydroxychalcone in licorice root extract, is regarded as a promising lead as a potential cancer chemopreventive agent 19, 20 ; . Isoliquiritigenin has previously been reported to suppress cyclooxygenase-2, known to play an important role in inflammation, and to induce apoptosis in mouse colon cancer cells 28 ; . The present study revealed that isoliquiritigenin blocked pro-inflammatory cytokine-induced protein expression of VCAM-1, E-selectin, and PECAM-1 at transcriptional levels and blunted THP-1 monocyte-endothelial cell interaction. The reduction in VCAM-1 noted in this study suggests an effect of isoliquiritigenin on endothelial function. It should be pointed out that co-culture of HUVEC with isoliquiritigenin and TNF-a did not cause toxicity data not shown ; , as determined by MTT assay. Nevertheless, a number of centres now offer a transplant service to such patients. Little information is available from these centres on how many patients with end-stage alcoholic liver disease have been excluded from their transplant programmes because of the presence of other alcohol-related disorders. Patients are rigorously evaluated before being accepted by transplant services, and it is to assumed that other organ damage is diligently sought, and, if identified and found to be clinically significant, would be a contraindication, relative or definite, to candidacy. Where transplantation has been undertaken in patients with alcoholic liver disease, outcomes are good, both in terms of survival and quality of life, and do not differ substantially from those reported in patients with non-alcoholic liver disease Bird et al., 1990; Kumar et al., 1990; Knechtle et al., 1992; Lucey et al., 1993 ; Fig. 3 ; . Importantly, it increases 2-year survival in the patients with the most severely decompensated disease Poynard et al., 1994 ; . Inhibitions approximating 40 per cent or less, results were not always consistent. This was noted in the tests involving urethan and purine hydrochloride with H.Ep. #3 and A-methopterin with H.S. #1, in which four discrepant tests were in cluded. DISCUSSION The availability of a method for growing human neoplasms in large numbers of laboratory animals made pertinent the possibility of their use in ex perimental chemotherapy screening systems. Two transplantable human tumors, the sarcoma H.S. #1 and the carcinoma H.Ep. #3, were selected for possible applicability to such purpose. It was recognized and appreciated that these neoplasms might not be representative of all types of human tumors, but their advantages were such as to lead to a determination of their suitability for experimental chemotherapy. Stock enumerated several criteria for assessing the usefulness of a tumor for experimental chem otherapy. These included rapid rate of growth, high percentage of takes, high degree of lethality, uniform growth, sensitivity to potential anti-tumor agents, and lack of response to the effects of inanition 17 ; . Data presented on most of these criteria indicate that the two human tumors, H.S. #1 and H.Ep. #3, satisfactorily fulfill many of these requirements. The rapid rates of growth of these two human transplantable tumors permitted evaluations to be made 9-11 days post-implantation for H.Ep. #3 and 11-12 days for H.S. #1. There were in dications that these termination points for ex periments could be reduced to the 7th day for H.Ep. #3 and to the 9th day for H.S. #1, if desirable. The degree of variability of tumor growth has always had an important influence on the evalua tion of a growth-inhibiting effect. The coefficients of variation of H.Ep. #3 and H.S. #1 tumor weights expressed the degrees of variability of these growths, that for H.Ep. #3 54-60 per cent ; being somewhat less than that for H.S. #1 72-73 per cent ; . The coefficients of variation for weight did not change to any great extent in the 12-month period illustrated in Table 2. In the West, we use it not only as a pre-eminent adaptogen, but also as an expectorant, immunomodulator, antiviral and anti-inflammatory. Licorice works specifically at the adrenal level, helping to alleviate adrenal insufficiency and thus promoting healthy HPA-axis function. Licorice also impacts the SAS, offering support in acute stress.18 In addition to licorice's impact on the nervous system via endocrine regulation, direct neurological effects have been suggested by laboratory investigations. Research using high doses equivalent to about 10 g in humans ; of an aqueous extract demonstrated a positive impact on learning and memory in mice, which may be due to anti-inflammatory and antioxidant properties, as well as facilitation of cholinergic transmission.22, 23 Further, recent research also suggested that the same dose of aqueous extract possesses an anti-depressant activity in mice, comparable to that of imipramine and fluoxetine.24 The investigators suggested regulation of norepinephrine and dopamine as a probable mode of activity. In keeping with Chinese tradition, I tend to use licorice in many formulas to harmonize the activity and flavor of the other herbs. I also find that the sweet and warming qualities of the plant give it a building nature, similar to that of food.
