The synthesis of mRNA and sRNA may be even less than the twofold inhibition observed. For example, in the sucrose fractionation experiment a relatively small breakdown of ribosomal RNA by nucleases would result in apparent inhibition of sRNA synthesis, because a given percentage of ribosomal RNA added to the 4S peak would provide a much larger increment of radioactivity in the control than in the inhibited culture. In the pulse-labeling and step-down experiments, some of the apparent inhibition of mRNA synthesis may result from the possibility that these procedures are not completely selective for the synthesis of mRNA alone. A particularly interesting situation exists in the experiments in which f3-galactosidase induction was measured in the presence of levorphanol. Mandelstam"3 has shown that the presence of a carbon source in a nongrowing culture leads to virtually complete catabolite repression of , 3-galactosidase induction. In our experiments there was induction at about half the normal rate in the presence of a carbon source in cultures not growing because of treatment with levorphanol. Even if levorphanol has no direct effect on , 3-galactosidase synthesis, the experiments of Mandelstam lead to the prediction that little or no induction should occur under these conditions because of catabolite repression. Our finding may be explained by the continued synthesis of DNA and protein which may result in the removal of catabolites responsible for repression. The recent report of Paigenl4 demonstrating that amino acids are potent repressors of f3-galactosidase synthesis supports this view. The decrease in rate of induction seen in our experiments may be a direct effect of the drug, or may be at least partially explainable by residual catabolite repression. The enzyme induction seen at concentrations of levorphanol which profoundly inhibit RNA synthesis gains added significance in the light of reports that other agents known to affect the synthesis of nucleic acids or proteins, such as actinomycin D, 15 mitomycin, 16 puromycin, 16 chloramphenicol, 14, 16 and streptomycin, 16 inhibit enzyme induction markedly at concentrations below those at which they exhibit measurable effects on macromolecular synthesis. The question may be raised whether the isotope incorporation into the 4S peak which persists in the presence of levorphanol may result from the fact that the enzyme which attaches the terminal trinucleotide ApCpC- ; to sRNA'7 18 is not inhibited. This has been found to be the explanation for the apparent resistance of sRNA in HeLa cells to actinomycin D.19 In the present experiments this possibility appears to be excluded. Since none of the three terminal bases contain methyl groups, the occurrence of considerable methylation 60-85%o of control ; of sRNA made in the presence of levorphanol during methionine starvation indicates that de novo synthesis of sRNA molecules has taken place. Moreover, paper electrophoresis of alkaline hydrolysates of sRNA isolated from cells labeled faith C'4-uracil in the presence of levorphanol showed that more than 50 per cent of the radioactivity was present in uracil.

Relative bioavailability is the comparison of absolute bioavailabilities of different forms of a chemical or for different exposure media containing the chemical US EPA 2005 ; . As direct measurement of bioavailability of soil-borne chemicals in humans is often impractical, various animal models in vivo * ; have been employed to study the bioavailability of toxic chemicals as a surrogate for children Freeman et al 1992; Casteel et al 1997; NEPI 2000; NRC 2003; US EPA 2005 ; Table 1 ; . The limitations of such an approach are that bioassays are specific to the test organism, chemical and matrix, therefore interpretation of data, and extrapolation to humans, needs careful consideration NRC 2003; Schoof 2004 ; . Moreover, these animal models have not been validated against estimates of bioavailability in humans for example, only one soil sample was studied for human bioavailability of lead, Maddaloni et al 1998 ; . Despite these limitations, the use of animal bioavailability assays has increased in the last two and half decades, particularly in the USA, in order to set clean-up goals for contaminated sites Casteel et al 1997; NRC 2003 ; . As routine use of animals in refining risk assessment is challenging in terms of cost, time, facility and expertise and ethical reservations, in vitro bioaccessibility methods attempting to mimic human gut conditions have been developed Ruby et al 1993, 1996; Drexler 1998; Sips 1998 ; . Bioaccessibility is a measure of dissolution of a contaminant from soil in an in vitro study, while bioavailability is the fraction that is absorbed in vivo. Therefore, the development of a reliable culture technique to test drug resistance or clinical validation of outcome measures related to genetic polymorphisms is needed. Culture of CMV in BAL was independently associated with an increased 2.7-fold ; risk of short-term mortality. The clinical significance of CMV in HIVrelated lung disease is an issue of controversy. Several studies3237 conducted before the introduction of adjunctive corticosteroid therapy concluded that CMV coinfection does not contribute directly to HIV-associated pulmonary disease. However, CMV does cause significant pulmonary disease in some individuals with HIV-1 infection, although the diagnosis frequently is missed antemortem.38, 39 It is possible that CMV may be a significant pathogen in patients treated with immunosuppressants or who have other infections. In one study, 40 the use of corticosteroids was related to the subsequent development of CMV retinitis and colitis in HIV-1 infected patients. In a previous study41 of PCP patients treated between 1989 and 1991, we showed a twofold increased mortality rate within 3 months of diagnosis in patients treated with adjunctive corticosteroids who were BAL CMV culture positive. However, the association did not reach statistical significance p 0.07 ; . A previous study42 found increased short-term mortality in patients with CMV isolated from BAL but did not report on use of corticosteroids. In the present study, a positive BAL CMV culture finding in patients receiving adjunctive corticosteroid therapy was, independently of CD4 count, associated with a statistically significant increased risk of death within 3 months of diagnosis. Patients not receiving corticosteroids who were BAL CMV negative were not at an increased risk. Further, autopsy material showed a high frequency of lung CMV inclusion bodies in patients who died within 3 months, but the number of patients precluded statistical conclusions. The mechanisms through which CMV may shorten survival of patients treated with corticosteroids are unknown. In hypothesis, corticosteroids may impair host defense mechanisms against CMV, or CMV may be a marker of an unidentified factor associated with severe disease. In vitro data suggest that CMV replication is increased in corticosteroid-treated macrophages.43 In contrast to our study, Azoulay et al11 recently found in a retrospective study that a Po2 60 mm Hg and BAL neutrophilia 10% predicted 90-day mortality among PCP patients managed from 1990 to 1995. In the present study, we applied the same statistical methodology but did not find these associations. There is no obvious explanation for this discrepancy because both study populations are comparable with regard to.

Abstract Late Variscan volcanic activity is documented in the Late Carboniferous Salvan-Dornaz sedimentary basin and in the neighboring basement units of the Aiguilles-Rouges and Mont-Blanc crystalline massifs Western Alps ; . Precise U Pb isotopic dating, zircon morphology and geochemical analyses indicate that volcanism occurred during shortlived pulses and that coexisting crustal and mantle sources were involved in the production of melts. Volcanic and subvolcanic products were emplaced along major NS to NNESSW transtensional fracture zones, similar to the ones that governed intense basement exhumation and that favored the formation and filling of the Late Carboniferous Salvan-Dornaz continental basin. In the Aiguilles-Rouges massif, dacitic flows outcropping at the base of the Salvan-Dornaz basin erupted at 308 3 Ma; they represent the surface equivalent of the nearby Vallorcine peraluminous granite and associated rhyolitic dykes 311 17 Ma ; . the Mont-Blanc massif, calc-alkaline rhyolitic dykes were emplaced simultaneously 307 2 Ma ; at shallow crustal levels, but they derive from deeper magma sources denoting enhanced mantellic activity. Recently identified tuffs and volcaniclastic layers embedded at different levels of the Salvan-Dornaz stratigraphic record testify a 295 + 3 4 old episode of highly explosive volcanism from distant volcanic centers, possibly located in the Aar-Gotthard massifs Central Alps ; . Their zircon typology is highly heterogeneous, documenting wall-rock contamination of the melts and or admixture of crustal sediments, whereas consistent subpopulations point to high-temperature magmas of deep-seated origin and alkaline affinity. The dated volcanic layers from the Salvan-Dornaz basin set the beginning of the detrital sedimentation at 308 3 Ma and constrain the deposition of 1.51.7 km thick of clastic sediments within a time span of 1015 Ma. These results infer minimum, long-term subsidence rates during basin evolution in the order of 0.1 mm a, while in the surrounding basement units estimated exhumation rates are in the range of 1 mm All dated rocks contain inherited zircon populations about 350, 450 or 600 Ma old.
